Last week, we had the honor to hold a seminar with National Biotechnology Research Park, Biomedical Translation Research Center, and Molecular Devices, to explore the potentials, future trends and revolution of organoids and organ-on-chips on new drug development. Hsian-Jean Chin, Director General, National Laboratory Animal Center, National Applied Research Laboratories, NARLabs said, those "human-based models" are not aim to totally replace animal models. In South Korea, pneumonia is a leading cause of death that continues to threaten public health every year. Through the tertiary hospital-based influenza surveillance system in South Korea, nasopharyngeal swab specimens were obtained from patients with unexplained cases of Community-Acquired Pneumonia (CAP) between 2011 and 2017. After real-time PCR screening using respiratory viral panels, the samples were found negative for 16 common respiratory pathogens including adenovirus, influenza viruses, rhinovirus, respiratory syncytial virus, coronavirus, metapneumovirus, and parainfluenza viruses. The aim of this study was to investigate the patient microbiota and examine the etiology of CAP requiring hospitalization. The nasopharyngeal microbiome of adult patients during CAP was analyzed using Next-Generation Sequencing (NGS) on the Illumina MiSeq platform and a subsequent bioinformatics pipeline. Efforts to combat the increasing threat of drug-resistant bacteria are being assisted by a new approach for streamlining the search for antimicrobial drug candidates, pioneered by researchers at Hokkaido University, led by Assistant Professor Kazuki Yamamoto and Professor Satoshi Ichikawa of the Faculty of Pharmaceutical Sciences. Their methods, developed together with researchers elsewhere in Japan and in the USA, are discussed in an article in the journal Nature Communications. The team split known Mray inhibitors into MraY binding regions (cores) and activity modulating regions (accessories). From 7 cores and 98 accessories, they generated a library of 686 MraY inhibitor analogs. These analogs were tested against MraY, and eight analogs possessing strong MraY inhibitory and antibacterial activity were identified. Various types of nucleic acids have been used as gene regulation tools or therapeutic agents for the treatment of human diseases with genetic disorders. Nucleic acids can also be used to develop sensing platforms for detecting ions, small molecules, proteins, and cells. Their performance can be improved through integration with other organic or inorganic nanomaterials. To further enhance their biological properties, various chemically modified nucleic acid analogues can be generated by modifying their phosphodiester backbone, sugar moiety, nucleobase, or combined sites. Alternatively, using nucleic acids as building blocks for self-assembly of highly ordered nanostructures would enhance their biological stability and cellular uptake efficiency. In this review, we will focus on the development and biomedical applications of structural and functional natural nucleic acids, as well as the chemically modified nucleic acid analogues over the past ten years. The recent progress in the development of functional nanomaterials based on self-assembled DNA-based platforms for gene regulation, biosensing, drug delivery, and therapy will also be presented. We will then summarize with a discussion on the advanced development of nucleic acid research, highlight some of the challenges faced and propose suggestions for further improvement.
